Age-associated nicotinamide adenine dinucleotide decline drives CAR-T cell failure
Chimeric antigen receptor (CAR) T cell therapy is one of the most promising cancer treatments. However, different hurdles are limiting its application and efficacy. In this context, how aging influences CAR-T cell outcomes is largely unknown. Here we show that CAR-T cells generated from aged female mice present a mitochondrial dysfunction derived from nicotinamide adenine dinucleotide (NAD) depletion that leads to poor stem-like properties and limited functionality in vivo. Moreover, human data...
mRNA metabolism regulator human antigen R (HuR) regulates age-related hearing loss in aged mice
Age-related hearing loss (ARHL) is among the most prevalent and complex disorders in older adults. However, the pathogenesis of ARHL remains poorly understood. Using a single-cell transcriptomic landscape of mouse cochlea at five time points (1, 2, 5, 12 and 15 months), we found that the levels of human antigen R (HuR)-a classical RNA-binding protein-increase with age. Here we show that HuR is specifically transported from the nucleus to the cytoplasm in hair cells in both aging mice and...
Endometrial aging is accompanied by H3K27ac and PGR loss
Whether and how endometrial aging affects fertility remains unclear. In our in-house clinical cohort at the Center for Reproductive Medicine of Peking University Third Hospital (n = 1,149), we observed adverse pregnancy outcomes in the middle-aged group after excluding aneuploid embryos, implying the negative impact of endometrial aging on fertility. To understand endometrial aging, we performed comprehensive transcriptomic profiling of the mid-secretory endometrium of young (<35 years) and...
A dynamic and multimodal framework to define microglial states
The widespread use of single-cell RNA sequencing has generated numerous purportedly distinct and novel subsets of microglia. Here, we challenge this fragmented paradigm by proposing that microglia exist along a continuum rather than as discrete entities. We identify a methodological over-reliance on computational clustering algorithms as the fundamental issue, with arbitrary cluster numbers being interpreted as biological reality. Evidence suggests that the observed transcriptional diversity...
Stepping through fear: analysing postural control in elderly women during transitional locomotor tasks
CONCLUSION: The study concludes that while FoF does not impair static balance, it significantly impacts movement initiation and execution in transitional tasks. Prolonged preparation time (S1) in the HIGH FoF group highlights the role of fear in delaying movement. These findings are important for designing interventions to reduce FoF and prevent falls in older adults.
Developing a comprehensive malnutrition prediction model for the elderly in nursing homes
CONCLUSION: This nomogram provides a robust tool for malnutrition risk stratification in nursing homes. Future studies should validate its generalizability across diverse populations and regions.
The association between stereoscopic vision and cognitive function on community-dwelling older adults: a cross-sectional study
CONCLUSIONS: Poor stereopsis was associated with cognitive dysfunction and had increased odds for cognitive impairment. Our findings suggest a cross-sectional association between stereopsis and cognitive dysfunction in community-dwelling older adults.
A monocentric prospective study investigating digital engagement among geriatric hospital patients
CONCLUSION: Our findings align with previous studies involving participants of comparable age, indicating comparable results, apart from disease-related participation restrictions. However, the significant proportion of patients who did not want to participate (55.7%) and the analysis of the reasons for nonparticipation suggest that the actual number of geriatric patients who do not engage online is higher. While this does not necessarily imply a complete rejection of digital products by this...
Assessing the impact of sleep quality on physical function in Chinese older inpatient
CONCLUSIONS: Poor sleep quality was associated with reduced physical function, especially in with advancing ageand in women. Targeted interventions to enhance sleep in the elderly may contribute to maintaining physical function and improve the quality of life of such patients.
A qualitative assessment of older adults' perspective on the utilization of information from preventive home visits
CONCLUSIONS: Older adults are a valuable resource for community planning aimed at promoting healthy aging. Further research is needed to explore the full potential of using information from preventive home visits for fostering inclusive age-friendly community planning.
Population size interacts with reproductive longevity to shape the germline mutation rate
Mutation rates vary across the tree of life by many orders of magnitude, with fewer mutations occurring each generation in species that reproduce quickly and maintain large effective population sizes. A compelling explanation is that large effective population sizes facilitate selection against weakly deleterious "mutator alleles" such as variants that modulate cell division or interfere with the molecular efficacy of DNA repair. However, while the fidelity of a single cell division largely...
Effects of Initiating Different Antidepressant Subclasses on Motor Vehicle Crash Risk Among Older Adults With Depression
CONCLUSION: We observed no differences in the one-year risk of MVC between antidepressant subclasses. When selecting among antidepressant subclasses to manage depression in older adults, MVC risk should not guide prescribing decisions, and other considerations should take precedence.
The Association Between Nutritional Status, Diet Quality, and Sleep Quality Among the Elderly in Jordan: A Cross-Sectional Study
Background: The percentage of the elderly population has increased worldwide. It has been estimated that environmental factors such as eating habits, sleep quality, and physical activity could be responsible for up to 75% of the aging process. Aims: To assess the nutritional status, diet quality, and sleep quality among the elderly in Jordanian. Methods: In a cross-sectional study targeting the Jordanian elderly, 426 participants agreed to participate. Sociodemographic data, anthropometric...
Sex-specific insights into drug-induced lifespan extension and weight loss in mice
The DrugAge database serves as a comprehensive resource for studying compounds that increase lifespan in model organisms. In the latest version of DrugAge, we implemented multiple updates, predominantly focusing on mouse studies to enhance data accuracy and consistency. Key improvements include re-recording of mouse data from original sources, standardization of drug dosages to parts per million, and recording of administration routes, treatment initiation ages, and lifespans of controls. The...
Targeting the chromatin remodeler BAZ2B mitigates hepatic senescence and MASH fibrosis
With increased age, the liver becomes more vulnerable to metabolic dysfunction-associated steatohepatitis (MASH) with fibrosis. Deciphering the complex interplay between aging, the emergence of senescent cells in the liver and MASH fibrosis is critical for developing treatments. Here we report an epigenetic mechanism that links liver aging to MASH fibrosis. We find that upregulation of the chromatin remodeler BAZ2B in a subpopulation of hepatocytes (HEPs) is linked to MASH pathology in patients....
Variation in albumin glycation rates in birds suggests resistance to relative hyperglycaemia rather than conformity to the pace of life syndrome hypothesis
The pace of life syndrome (POLS) hypothesis suggests that organisms' life history and physiological and behavioural traits should co-evolve. In this framework, how glycaemia (i.e. blood glucose levels) and its reaction with proteins and other compounds (i.e. glycation) covary with life history traits remain relatively under-investigated, despite the well-documented consequences of glucose and glycation on ageing, and therefore potentially on life history evolution. Birds are particularly...
Tooth Loss and 12-Year Mortality Risk in 8494 Older Adults From Ireland
CONCLUSIONS: Edentulism was independently associated with all-cause mortality in a cohort of 8494 men and women from Ireland. Edentulism was significantly associated with respiratory and cancer mortality.
Immunosenescence Profile Is Associated With Increased Susceptibility to Severe COVID-19
In this study, we tested the hypothesis that the immunosenescence profile could account for the disproportional susceptibility of the elderly to severe forms of COVID-19. The immunological profiles of volunteers residing in endemic and non-endemic areas for chronic infectious diseases were analyzed at the early stage of SARS-CoV-2 infection. A unique signature of inflammatory plasma mediators was identified in COVID-19 volunteers when compared to individuals with other flu-like syndromes....
Beyond Longevity: Prioritizing Individualized Care in Older Adults With NSTE-ACS
Four domains guide management choice: geriatric assessment, cardiovascular risk-factor assessment, medical therapy optimization, and angiographic considerations, integrated to tailor management strategies among older adults with NSTE-ACS.
VPS33B-dependent exosomes modulate cellular senescence of mesenchymal stem cells via an autocrine signaling pathway
Mesenchymal stem cell (MSC)-derived exosomes have been intensively studied for their therapeutic effects on tissue repair and regeneration. However, the specific contributions of exosomes derived from endogenous bone marrow MSCs to the maintenance of bone tissue homeostasis remain unclear. In this study, we impaired MSC-derived exosome secretion by specifically deleting vascular protein sorting 33B (VPS33B). Mice deficient in VPS33B (VPS33B-cKO mice) exhibited premature bone loss and imbalanced...
