Periarteriolar niches become inflamed in aging bone marrow, remodeling the stromal microenvironment and depleting lymphoid progenitors
In early postnatal and young adult bone marrow, Leptin receptor-expressing (LepR^(+)) stromal cells and endothelial cells synthesize factors required for hematopoietic stem cell (HSC) maintenance, including Stem Cell Factor (SCF) and Cxcl12. However, little is known about how these stromal cells change during aging. We performed single-cell RNA sequencing of mouse bone marrow stromal cells at 2, 12, and 24 mo of age. We identified five transcriptionally distinct subsets of LepR^(+) cells, all of...
Serum Proteomic and Metabolomic Signatures of High Versus Low Physical Function in Octogenarians
Physical function declines with aging, yet there is considerable heterogeneity, with some individuals declining very slowly while others experience accelerated functional decline. To gain insight into mechanisms promoting high physical function with aging, we performed proteomics, targeted metabolomics, and targeted kynurenine-focused metabolomic analyses on serum specimens from three groups of octogenarians: High-functioning master athletes (HF, n = 16), healthy normal-functioning non-athletes...
Impacts of ageing on the efficacy of CAR-T cell therapy
Chimeric antigen receptor T cells recognizing CD19 (19CAR-T) cell therapy has achieved robust clinical efficacy when treating some hematological malignancies, but which patient subgroups benefit mostly remains elusive. Here we summarized the data of 541 patients from 30 clinical trials who underwent 19 CAR-T therapy and analyzed the different clinical responses between young (<44 years), middle-aged (45-59 years) and elderly (>60 years) patients and found that the young patients showed a higher...
Latest developments of microphysiological systems (MPS) in aging-related and geriatric diseases research: A review
Aging is a gradual and irreversible process accompanied by the decline in tissue function and a significantly increased risk of various aging-related and geriatric diseases. Especially in the paradoxical context of accelerated global aging and the widespread emergence of pandemics, aging-related and geriatric diseases have become leading causes of individual mortality and disability, drawing increasing attention from researchers and investors alike. Despite the utility of current in vitro...
Hippocampal Sclerosis: A Review on Current Research Status and Its Mechanisms
Hippocampal sclerosis (HS) is a pathological condition characterized by significant loss of hippocampal neurons and gliosis. This condition represents the most common neuropathological change observed in patients with temporal lobe epilepsy (TLE) and is also found in aging individuals. TLE related to HS is the most prevalent type of drug-resistant epilepsy in adults, and its underlying mechanisms are not yet fully understood. Therefore, developing improved methods for predicting and treating...
Racial and Ethnic Disparities in Accessing High-Quality Home Health Care Among Older Adults With and Without Dementia
CONCLUSIONS AND IMPLICATIONS: Racial and ethnic minority individuals face significant disadvantages in accessing high-quality HHC, with persons living with dementia from these groups being the most disadvantaged. Further research is warranted to investigate the referral and admission processes for HHC. Our findings highlight the need for actions from clinicians and policymakers to tackle potential biases in the aforementioned care processes.
Temporal Changes in Alzheimer's Disease-Related Biomarkers in the CSF of Cognitively Normal Subjects at Different Ages: The Chongqing Ageing and Dementia Study
Revealing the temporal evolution of cerebrospinal fluid (CSF) biomarkers during aging is critical to understanding disease pathogenesis and developing early diagnoses and interventions for Alzheimer's disease (AD). CSF was obtained from 549 cognitively normal subjects between 18 and 93 years of age. 12 AD-related biomarkers were evaluated, including amyloid β (Aβ42, Aβ40, Aβ42/Aβ40 ratio), hyperphosphorylated tau (P-tau), neuronal injury/degeneration (T-tau, NFL, NSE, H-FABP, VILIP-1),...
The pro-inflammatory cytokine IL6 suppresses mitochondrial function via the gp130-JAK1/STAT1/3-HIF1α/ERRα axis
Chronic inflammation and a decline in mitochondrial function are hallmarks of aging. Here, we show that the two mechanisms may be linked. We found that interleukin-6 (IL6) suppresses mitochondrial function in settings where PGC1 (both PGC1α and PGC1β) expression is low. This suppression is mediated by the JAK1/STAT1/3 axis, which activates HIF1α through non-canonical mechanisms involving upregulation of HIF1A and ERRα transcription, and subsequent stabilization of the HIF1A protein by ERRα....
Modular mechanisms of immune priming and growth inhibition mediated by plant effector-triggered immunity
Excessive activation of effector-triggered immunity (ETI) in plants inhibits plant growth and activates cell death. ETI mediated by intracellular Toll/interleukin-1 receptor/resistance protein (TIR) nucleotide-binding, leucine-rich repeat receptors (NLRs) involves two partially redundant signaling nodes in Arabidopsis, ENHANCED DISEASE SUSCEPTIBILITY 1-PHYTOALEXIN DEFICIENT 4-ACTIVATED DISEASE RESISTANCE 1 (EDS1-PAD4-ADR1) and EDS1-SENESCENCE-ASSOCIATED GENE 101-N REQUIREMENT GENE 1...

Systemic immune inflammation mediates the association of serum omega-3 and omega-6 polyunsaturated fatty acids with biological aging: a national population-based study
CONCLUSIONS: This study demonstrated that higher levels of certain PUFAs were associated with a reduction in PhenoAge acceleration either individually or in combination, with DHA having the most prominent effect in mixed effects. The SII mediated these relationships, suggesting that PUFAs may slow biological aging by reducing inflammation. These findings highlighted the potential role of PUFAs in mitigating accelerated aging and their implications for aging-related health interventions.
Repositioning coal power to accelerate net-zero transition of China's power system
Countries are urged to advance the energy transition in a just, orderly, and equitable manner, yet the appropriate pathway remains unclear. Using a provincial-level, hourly-dispatched power system model of China that incorporates intertemporal decisions on early retirement and carbon capture retrofitting, our study reveals that for coal-rich but gas-poor economies, repositioning coal power from a baseload resource to a flexibility provider can accelerate net-zero transition of the power system...
Associations between the oral microbiome, number of teeth and frailty among American adults: A cross-sectional study from NHANES 2009-2012
CONCLUSION: Lower oral microbiome diversity is associated with higher frailty and mortality. The number of teeth partially mediates this link, emphasizing the importance of oral health in mitigating frailty and promoting healthy aging.
Mechanistic insights into stem cell fate regulation via RNA methylation
Stem cells possess an extraordinary ability for self-renewal and differentiation, making them essential for tissue repair, regeneration, and anti-aging. RNA methylation is crucial in regulating stem cell fate by modulating gene expression. This review synthesizes current research on RNA methylation modifications, such as m⁶A, m⁷G, m⁵C, and m¹A, and their impact on adult stem cell fate. It provides a comprehensive overview of the molecular machinery involved in RNA methylation, emphasizes the...

Fetal-like reversion in the regenerating intestine is regulated by mesenchymal asporin
Mesenchymal cells and the extracellular matrix (ECM) support epithelium during homeostasis and regeneration. However, the role of the mesenchyme in epithelial conversion into a fetal-like regenerative state after damage is not known. We modeled epithelial regeneration by culturing intestinal epithelium on decellularized small intestinal scaffolds (iECM) and identify asporin (Aspn), an ECM-bound proteoglycan, as a critical mediator of epithelial fetal-like reprogramming. After damage, transient...
COVID-19 booster doses reduce sex disparities in antibody responses among nursing home residents
CONCLUSIONS: The blunting of sex differences in antibody response observed following the primary series by the 1st booster dose underscores the importance of booster vaccination in this population.
Age-related differences across the adult lifespan: a comparison of six field assessments of physical function
CONCLUSIONS: These findings imply that most field tests are responsive to age-related declines in physical and/or muscle function after 60. In younger age groups, field tests that evaluate lower-limb power and have minimal ceiling effects, such as SCP and CMJ, should be prioritized.
Continuous map of early hematopoietic stem cell differentiation across human lifetime
Uncovering early gene network changes of human hematopoietic stem cells (HSCs) leading to differentiation induction is of utmost importance for therapeutic manipulation. We employed single cell proteo-transcriptomic sequencing to FACS-enriched bone marrow hematopoietic stem and progenitor cells (HSPCs) from 15 healthy donors. Pseudotime analysis reveals four major differentiation trajectories, which remain consistent upon aging, with an early branching point into megakaryocyte-erythroid...
